Product Description
Bupivacaine hydrochloride is an amide type local anesthetic commonly used for nerve block anesthesia, local infiltration anesthesia, and epidural anesthesia. Its mechanism of action is similar to that of amide type local anesthetics, in which the drug first binds to plasma proteins, metabolizes through the liver, and is then excreted from the urine through the kidneys.
Its characteristics are: (1) strong anesthetic power (about 4 times that of lidocaine); (2) The duration is relatively long (usually around 5 hours, with the longest reaching more than 10 hours); (3) Has minimal impact on the respiratory and circulatory systems; (4) Pregnant women using this product are not prone to spreading and affecting the fetus. This product is already a relatively mature product abroad (included in the 1973 edition of the British Pharmacopoeia), and there are abundant literature reports on its synthesis. Many domestic literature reports also provide detailed descriptions of its synthesis route. The present invention mainly relates to the final step of the alkylation reaction and the salt formation reaction. According to literature reports, most of the alkylation reactions are carried out in the solvent DMF. After the reaction is completed, it is added to ice water to obtain bupivacaine base. The salt formation reaction is to dissolve bupivacaine base in isopropanol, then pass hydrogen chloride gas and filter to obtain bupivacaine hydrochloride. Alternatively, after the reaction is completed in DMF, dilute hydrochloric acid can be directly added and cooled to crystallize to obtain bupivacaine hydrochloride. The disadvantages of the preparation methods reported in existing literature are the large amount of DMF used, difficult recovery, and high production costs.
